The main objective of the CAMP project (2009-2011) was to estimate the effective connectivity between Marine Protected Areas (MPA) in the southwest Indian Ocean (SWIO) through population genetics of 3 biological models Epinephelus merra, Lutjanus kasmira and Myripristis berndti. The ultimate goal is to provide scientific results for managers to contribute significantly to the design of the MPA network in SWIO. The main aim of the CAMP project (2009-2011) is to estimate the efficient connectivity between the Marine Protected Areas (MPA) of the South-West Indian Ocean (SWIO). For that purpose, we use a population genetics approach on 3 reef fishes: Epinephelus merra, Lutjanus kasmira et Myripristis berndti. The overall goal of the project is to provide new elements to MPA managers that will contribute to design the best network of MPAs in the SWIO. In the first year of the project, most of the time was spent to sample fishes. Characterisation of 22 polymorphic microsatellite loci for the Broadbill Swordfish, Xiphias gladius ArchiMer

The Broadbill Swordfish is harvested by fisheries throughout the world. In order to devise more effective management strategies, we need a clear understanding of the population structure of the species. From a library of 29 tetranucleotide repeats, 22 microsatellite markers were characterised for 94 swordfish samples captured from eastern and western Australia. The number of alleles ranged from 2 to 26 and observed heterozygosity from 0.066 to 0.923. We have identified 18 microsatellites that will be valuable in the examination of swordfish population structure.

Characterization of thirteen new polymorphic microsatellite markers from the honeycomb grouper Epinephelus merra ArchiMer

Epinephelus merra (Serranidae) is the most common and widespread species of this genus, mostly found in reef habitats from South Africa to Central Pacific. Thirteen polymorphic microsatellite loci were isolated and characterized from this species. Polymorphism was studied in two localities from the Comoros archipelago-Moh,li and Moroni. Allelic diversity was 7.5, polymorphism ranging from 3 to 13 alleles. Overall heterozygosity was high with an average observed heterozygosity of 0.587. Three of these loci were characterised by heterozygote deficiencies for both populations. Finally, ten of the 13 developed primers could be used in future population genetics study of E.merra that might be useful in a context of marine biodiversity conservation.

Comparative phylogeography of the western Indian Ocean reef fauna ArchiMer

Assessing patterns of connectivity at the community and population levels is relevant to marine resource management and conservation. The present study reviews this issue with a focus on the western Indian Ocean (WIO) biogeographic province. This part of the Indian Ocean holds more species than expected from current models of global reef fish species richness. In this study, checklists of reef fish species were examined to determine levels of endemism in each of 10 biogeographic provinces of the Indian Ocean. Results showed that the number of endemic species was higher in the WIO than in any other region of the Indian Ocean. Genetic connectivity of the reef fish Lutjanus kasmira at the scale of the western Indian Ocean ArchiMer

Examining the genetic structure of species allows an estimate of the level of evolutionary connectivity between localities; this information is important for marine biodiversity protection, in particular, for the delineation of marine protected areas. In this context, a total of 601 Lutjanus kasmira (Forsskal, 1775) were sampled in 16 localities of the western Indian Ocean and analyzed with both mitochondrial cytochrome b sequencing and eight microsatellite loci genotyping. Both genetic markers indicate that differentiation was not significant even between samples separated by more than 4000 km. Genetic population structure of the Swordfish (Xiphias gladius) in the southwest Indian Ocean: Sex-biased differentiation, congruency between markers and its incidence in a way of stock assessment ArchiMer

Genetic variation was surveyed at 11 microsatellite loci and at 517 bp of the mitochondrial control region to investigate the presence of genetic stock structure in swordfish (Xiphias gladius) in four proximal localities of the southwest Indian Ocean. One aim of this study was to serve as a preliminary examination for congruency of structure detected by these two genetic markers, prior to conducting a more comprehensive basin-wide survey of the Indian Ocean and nearby surrounding areas. Genetic structure of the reef grouper Epinephelus merra in the West Indian Ocean appears congruent with biogeographic and oceanographic boundaries ArchiMer

The reef fauna connectivity of the West Indian Ocean (WIO) is one of the least studied globally. Here we use genetic analyses of the grouper Epinephelus merra (Bloch 1793) to determine patterns of connectivity and to identify barriers to dispersal in this WIO marine area. Phylogeographic and population-level analyses were conducted on cytochrome b sequences and microsatellites (13 loci) from 557 individuals sampled in 15 localities distributed across the West Indian Ocean. This article documents the addition of 92 microsatellite marker loci to the Molecular Ecology Resources Database. These loci were cross-tested on the following species: Calephelis arizonensi, Calephelis borealis, Calephelis nemesis, Calephelis virginiensis and Lutjanus bengalensis.

Genetic population structure of swordfish Xiphias gladius was examined based on 2231 individual samples, collected mainly between 2009 and 2010, among three major sampling areas within the Indian Ocean (IO; twelve distinct sites), Atlantic (two sites) and Pacific (one site) Oceans using analysis of nineteen microsatellite loci (n = 2146) and mitochondrial ND2 sequences (n = 2001) data. Population structure enhances perspectives on regional management of the western Indian Ocean green turtle ArchiMer

To refine our understanding of the spatial structure of the green turtle (Chelonia mydas) populations in the South West Indian Ocean (SWIO), we analysed patterns of mitochondrial DNA (396 base pairs control region fragment) variation among 171 samples collected at five distinct locations (Kenya, Northern Mozambique, and three locations in the Republic of Seychelles: the Granitic, Amirantes, and Farquhar groups) and compared them to genetic data (n = 288), previously collected from 10 southern locations in the SWIO. Restricted dispersal of the reef fish Myripristis berndti at the scale of the SW Indian Ocean ArchiMer

The reef fish Myripristis berndti (Jordan & Everman 1903) is a pantropical species. A genetic analysis was conducted on 353 individuals from 10 localities distributed across the SW Indian Ocean (SWIO) in order to determine patterns of connectivity in the SWIO. Both the mtDNA sequences (711-bp cytochrome b sequences) and the microsatellites (8 newly developed loci) reveal spatial patterns of differentiation within the SWIO. There is, however, a discrepancy between the structure observed with each kind of marker.
